Temp doesn't matter, as long as you can make sure the stuff in the bottle you're using is the right temp. The piss that comes out of you into the toilet (don't let a lot in but a bit) will cool down significantly once it hits the water, I don't really believe it's possible to get an accurate reading on the temp in the toilet vs the temp of the sample as you can sit in the bathroom for a few mins after you fill the cup and just say you were finishing up. Not that I believe they will ask. But make sure you hide that container somewhere other than a pocket or someplace visible. Every test I've ever taken they make you remove everything from your pockets (up and including turning out pockets to PROVE there is nothing in them).
