do some column chromatography on the extracted dye. verify by NMR/MS/GC(or whatever) how pure your sample is

the dye is red it will look crazy awesome when you run the column.

in grad school i did several columns every day for months so i'm not sure if it will be hard for you to learn because i can do them while sleeping

Should be able to learn in a few days by watching YouTube videos but there's a lot of variables you'll need to know sir.

I don't imagine size exclusion would work too well on a small molecule and it's not hydrophobic and as far as I can tell doesn't have any metal to do metal affinity so you'll want to do ion exchange chromatography.

Basically you'll need to find the pI of your anthocyanin. pI is the point at which the molecule has a neutral charge, so if your buffer has a pH higher than the pI of your anthocyanin your anthocyanin will be negatively charged and vice versa.

Next you create an ion exchange resin, which is a bunch of tiny beads that are either cation exchangers or anion exchangers. Suspend in a buffer, pack your column, load your sample and elute with a salt gradient.

Note that if your school doesn't have the equipment it will be prohibitively expensive. Even the resin is like $500/bottle depending on what type you use.

Like I said go practice on Apps

It's designed to teach chromatography techniques for proteins but will work well for this as well.

just do gravity column its more easier. buy some sand and silica

Alrighty thanks for the tips guys.

What ill do 1st is just do some extracts, wont worry bout purity initially, most important thing right now I think is to ensure i can build a working cell..

Then when ive done that i will be free of that anxiety and can enjoy making "chromo columns"

Enjoy might be a strong word at first. Programming the column computers, while not overly difficult, can be very frustrating til you get the hang of it.

Wait is column chromatography also a means of doing a very pure extraction?

I saw a vid on youtube describing Anthocyanin extraction using that method

"10–12 fresh raspberry fruits were collected and dip coated in 20ml
water for one day. The aqueous solution was then filtered to get a clear
red liquid which is the anthocyanin dye without further purification."

keep it simple

after doing my masters at a badly underfunded university i became a lot more sensitive to how much stuff costs. do everything you can to avoid paying high prices. steal articles from the internet. pirate software. if some chemical you need is expensive try making it. avoid using expensive machines until its no longer avoidable

I submitted my order request of FTO glass today.

Getting 100 pieces of 1cm x 1cm FTO glass (pretty small I know, but the spectrometer I'm using has a small holding compartment and I think 15mm is the biggest I can go anyway) from an american based company.. for $200 USD.. whatever that is Canadian. But that means up to 100 DSSCs

And on Wednesday I'm going to the chemistry store with my prof to see if I can acquire all the chemicals I need.

I just remembered when I was 11 I was at a farm with my sister and her friend and her friend's family.


I mentioned to the friend's mom when we were talking alone that I had seen something at school that had to do with sex. She told me she wanted me to tell her and her friend what I saw, but not now, later.


So later after dinner, the mom and the mom's friend and me were sitting in the barn alone together and they asked me to tell them what I saw at school that had to do with sex. I wasn't in trouble, no. I knew they wanted to hear it because they wanted to hear it...





I sat there surrounded by wet cougar minge as I recounted a dirty limerick I saw on a locker door.






And that was my first experience disappointing a woman.
